Genetic Analyses of Heat-Sensitve Pawn Mutants of PARAMECIUM AURELIA.

چکیده
Heat-sensitive Pawn (ts Pawns) of Paramecium aurelia behaved normally when grown at 23 degrees but failed to avoid strong stimuli at 35 degrees . Four of the five ts Pawn lines tested were found to be allelic at a locus known also to carry temperature-independent Pawn mutations. The fifth ts Pawn line complemented all the conditional and unconditional Pawn mutants of the two known loci. This result, together with the patterns of F(2) segregation from various crosses, suggested the existence of a third Pawn locus. An additive effect of the unlinked ts Pawn genes was observed. These findings and the significance of ts Pawns as experimental material in behavioral and physiological research are discussed.

Calcium channel stability measured by gradual loss of excitability in pawn mutants of Paramecium aurelia.
Mutants of Paramecium aurelia that are unable to reverse swimming direction are called pawns. They lack the inward ionic (calcium) current required for the upstroke of the electrically excitable membrane response. By following the progressive loss of reversal response and excitability in cells that are suddenly changed from a heterozygous (wild-type) state to a homozygous mutant state, an estim...
متن کاملMicroinjection of cytoplasm as a test of complementation in Paramecium
Mutants in Paramecium tetraurelia, unable to generate action potentials, have been isolated as cells which show no backward swimming in response to ionic stimulation. These "pawn" mutants belong to at least three complementation groups designated pwA, pwB, and pwC. We have found that microinjection of cytoplasm from a wild-type donor into a pawn recipient of any of the three complementation gro...

The reversal response in Paramecium aurelia is mediated by calcium which carries the inward current during excitation. Electrophysiological studies indicate that strontium and barium can also carry the inward current. Exposure to high concentrations of barium rapidly paralyzes and later kills wild-type paramecia. Following mutagenesis with nitrosoguanidine, seven mutants which continued to swim...
متن کاملThe cloning by complementation of the pawn-A gene in Paramecium.
The genetic dissection of a simple avoidance reaction behavior in Paramecium tetraurelia has shown that ion channels are a critical molecular element in signal transduction. Pawn mutants, for example, were originally selected for their inability to swim backward, a trait that has since been shown to result from the loss of a voltage-dependent calcium current. The several genes defined by this p...
متن کاملBehavioral Mutants in PARAMECIUM CAUDATUM.
Mutants of Paramecium caudatum with abnormal swimming behavior or responses to cations were obtained by mutagenesis with N-methyl-N'-nitro-N-nitrosoguanidine. Some of the mutants, like pawn in P. tetraurelia, cannot swim backward and are called CNR. Seven independently obtained CNR mutants belonged to three complementation groups, designated as cnrA, cnrB and cnrC. Some characteristics of doubl...
